Abstract

Piles of plastic waste at the Piyungan Landfill in Bantul Yogyakarta can be a source of microplastics and endanger if they accumulate in the bodies of living things. Data collection on microplastic contamination in leachate ponds at the Piyungan landfill is essential because the leachate will go to the Opak River, which is still used by the community. This study aimed to analyze the abundance and polymer types of microplastics in the leachate at the Piyungan landfill. The study was conducted from January to April 2021. Leachate samples were taken at the inlet pond (before treatment) and outlet pond (after treatment). The research stages include leachate sampling, filtering particles, drying samples, density separation, separation with organic compounds, and filtering supernatants containing microplastics. Identification of microplastics was carried out visually using a microscope, while the identification of polymer types was determined by the Fourier-transform infrared streptoscopy test. The data were analyzed quantitative descriptively, and inferentially to compare the average abundance value and determine the correlation between microplastic abundance and abiotic parameters. The analysis results showed the abundance of microplastics in the inlet pool of 154.8 ± 21.22 particles/L while in the outlet pool of 135.60 ± 12.18 particles/L. According to the T-paired test, it was found that the abundance of microplastics in the two ponds was not significantly different. Water temperature correlates with the abundance of microplastic in the leachate. The identified polymer types are Polystyrene (PS), Polyethylene (PE), and Polyvinyl Chloride (PVC), which are commonly used as raw materials for single-use plastic, food containers, and leachate sewer pipes. This study concludes that in the leachate at the Piyungan landfill, the highest microplastic was found in the inlet pond with polymer types, namely Polystyrene (PS), Polyethylene (PE), and Polyvinyl Chloride (PVC). Therefore, the Yogyakarta government needs to increase the leachate processing process to decompose this microplastic and not pollute the Opak river as a leachate disposal channel for the Piyungan landfill.

Abstract

Sidoarjo is a district that has a mangrove ecosystem, but due to the rapid development in Sidoarjo, there is a lot of conversion of mangrove land into residential land. Therefore, it is necessary to collect data on mangroves in the Coastal of Sedati District and Jabon District, Sidoarjo Regency. The purpose of this study was to determine the index of diversity value and the index of importance of mangroves on the coast of Sedati and Jabon sub-districts, Sidoarjo district, with a trans-sect line plot of 10×10 along 300 meters perpendicular to the shoreline at 2 predetermined observation stations. Observational data were analyzed with the Shannon-Weiner diversity value index and the significance value index. Based on observational data, the diversity index obtained is 0,942 and the value of H' is less than 1, indicating that the diversity index on the coast of Sedati District and Jabon District, Sidoarjo Regency is low. The important value index on the coast of Sedati District and Jabon District, Sidoarjo Regency is 298.058% for trees and 22.02% for seedlings, and Avicennia marina has the highest important value index with a value of 198.25. Environmental conditions at station 1 in Sedati District showed unfavorable environmental conditions for the distribution of various types due to the high thickness of the mud, causing some species to have difficulty adapting, but at observation station 2 in Jabon District it supported the distribution because it had a medium to thin mud thickness so that various species adaptable.

Abstract

The increase of freshwater aquaculture leads to the increasing demand for silkworms (Tubifex sp) as natural feed. Unfortunately, high demand of silkworm supply is not fully fulfilled yet because its availability is relied on natural catch. Cultivation of Tubifex sp. outside its natural habitats requires good information on substrates and feed. We aimed to study the effect of  mud, sand and their combination use as substrates in supporting Tubifex sp growth. This study used complete randomized design with 5 treatments, namely K1 (100% sand), K2 (100% mud), P1 (combination 75% mud-25%) sand, P2 (combination 50% mud-50% sand), and P3 (combination 25% mud-75% sand) with triplicates on each treatment. Research was conducted for 21 days using fermented chicken manure as feed. Tubifex sp growth was measured on biomass, number of individuals, and average body weight, while pH, temperature, dissolved oxygen and total organic matter were listed as measured environmental parameters. The use of sand and mud combination as substrate was better than a single sand or mud alone to support Tubifex sp growth. Combination of 75% mud and 25% sand showed the best growth of Tubifex sp as observed on biomass value of 13,244 g, 370 number of individuals, and average weight body of 2,741 mg. The application of a combination of sand and mud substrates has a potency to support the cultivation of Tubifex sp outside its native habitat.

Abstract

Brown algae is a marine functional food that high fibers, vitamins, and bioactive compounds. Brown algae improved blood glucose and lipid profile. This study investigated the potential activity of two bioactive compounds from brown algae, as anti-obesity through pancreatic lipase inhibition. Molecular docking was conducted for investigating the lipase inhibition of bioactive compounds. Lipase structure (PDB ID 1ETH) was obtained from protein data bank and docked with fucodiphlorethol and phloroglucinol in specific position. Then, the complex was analyzed by PyMol following by Discovery Studio version 21.1.1. Phloroglucinol - pancreatic lipase showed five amino acid residues by hydrogen bonds, hydrophobic and electrostatic.  Fucodiphlorethol - pancreatic lipase generated binding energy -392 kJ/mol and consisted of eight hydrogen bonds, three electrostatic, and 4 hydrophobic interactions. The active sites of fucodiphlorethol and phloroglucinol with lipase were not considered as active sites or catalytic sites. Therefore, we predicted that both of fucodiphlorethol and phloroglucinol inhibit lipase allosterically by altering the protein structure conformations. molecular docking analysis suggested that fucodiphlorethol and phloroglucinol have potential antiobesity effect by inhibiting lipase. Molecular dynamic are needed for further investigation.
